The Protein Engineering and In Vitro Discovery Team within Biologics Discovery is responsible for the design of lead biologic candidates. In this position, the Senior Scientist will be an integral member of the Multispecific Antibody Engineering Group that works on the cutting edge of technology to deliver "therapeutic design fit-for-purpose" into the pipeline. The position offers an exciting opportunity to not only engage in the discovery and molecular engineering of biotherapeutics, but also develop new processes and technologies. This role is focused on the design and development of multispecific antibodies. This Senior Scientist will have the opportunity to lead project teams and interact seamlessly with world-class Therapeutic Area scientists.

Key Responsibilities:

  • Lead/participate on project teams to advance discovery portfolio projects.
  • Champion cross-functional technology development efforts aimed at fulfilling strategic long-term antibody design initiatives and validation.
  • Independently present/defend scientific findings in multi-functional project teams/initiatives.
  • Independently plan, analyze and interpret experimental results and draft patent applications, manuscripts, protocols, SOPs, technical reports, etc.


Qualifications

Education:

  • Minimum of Ph. D. in structural biology, biochemistry, or biophysics with a minimum 2 years of experience post-Ph. D. is required.

Experience and Skills:

Required:

  • In-depth knowledge and demonstrated experience in protein structure and analysis, including structure determination methods (e.g. x-ray crystallography, cryo-electron microscopy, NMR) and structure-based protein design is required.
  • Sound understanding of biophysical and biochemical methods for protein analysis, including ELISA, SPR, and/or flow cytometry, thermal stability analysis, and protein expression/purification from mammalian and bacterial cells is required.
  • Expertise in protein structure modeling and analysis (e.g. Schrodinger, MOE, pyMol, Chimera, etc.) is required.
  • Detailed knowledge of molecular biology, including expression vector design, protein expression from mammalian host cells and purification (affinity chromatography, ion-exchange chromatography, etc) is required.
  • Zeal for addressing complex scientific questions with creativity and a collaborative spirit. Demonstration of successful implementation of new ideas with calculated risk taking. Abilities to lead by influence and communicate across teams with diverse backgrounds is required.
  • Demonstration of clear, concise, and timely communication skills, exemplified by peer-reviewed publications is required.

Preferred:

  • A minimum of 1 year of industry experience is preferred.
  • Expertise in scientific computing (e.g. Python) and knowledge of in silico protein structure analysis is highly preferred.
  • Detailed understanding of antibody sequence analysis, including post-translational modification risks, germline analysis, and Fc-mediated effector functions is preferred.
  • Experience with of the state-of-the-art molecular biology and protein engineering methods, including antibody and cytokine is preferred.
  • Understanding of antibody and cytokine structure and function, including modulation of target binding and Fc-based effector function is preferred.
  • Hands on experience in HTP-based binding assays (such as ELISA, SPR, MSD) and/or cell-based (Intellicyt/iQue) for screening antibody variants is preferred.
